Drug developers are broadening their focus beyond single-acting GLP-1 agonists (drugs targeting a weight-regulating hormone pathway) toward dual and triple agonists, amylin-based medicines, oral treatments and long-acting formulations. Novo Nordisk's Wegovy, approved by the FDA (U.S. medicines regulator) in June 2021, helped establish the modern obesity-drug market, while Eli Lilly's Mounjaro, which targets both GLP-1 and GIP, demonstrated stronger weight-loss effects and intensified competition around multi-acting treatments. Triple agonists adding glucagon are viewed as potential next-generation drugs, while Celltrion and Daewon Pharmaceutical are developing quadruple agonists. Hanmi Pharmaceutical's HM17321, licensed to Genentech, uses UCN2 rather than GLP-1 and is designed to reduce fat while building muscle. Other companies are testing amylin mimics, muscle-preserving agents and mechanisms differentiated from GLP-1. Developers are also working on oral, once-monthly and once-weekly delivery options to address injection burdens and adherence. Researchers say future market leadership is likely to depend on a balance of long-term safety, the quality of weight loss, dosing convenience, manufacturing stability and insurance access, rather than weight reduction alone.
