Rigel Pharmaceuticals said VEPPANU (vepdegestrant) is now available by prescription in the United States for adults with ER-positive, HER2-negative, ESR1-mutated advanced or metastatic breast cancer, as detected by an FDA-authorized test, after disease progression following at least one line of endocrine therapy. The 200 mg once-daily oral drug, distributed through specialty channels in the United States and Puerto Rico and priced at $29,400 for a 30-day supply, is the first and only FDA-approved PROTAC. Rigel said a pivotal trial showed statistically significant and clinically meaningful progression-free survival improvement versus fulvestrant and that VEPPANU was generally well tolerated, while the label warns of QTc prolongation, embryo-fetal toxicity and other adverse reactions.
