Johnson & Johnson reported new results from two studies of RYBREVANT in advanced non-small cell lung cancer (NSCLC). In the Phase 2b COPERNICUS study, 214 U.S. patients with previously untreated advanced NSCLC carrying common EGFR mutations received subcutaneous RYBREVANT FASPRO (amivantamab and hyaluronidase-lpuj) plus LAZCLUZE (lazertinib) alongside preventive measures. After a median follow-up of 8.3 months, rash occurred in 25% of patients, while administration-related reactions and venous thromboembolism each occurred in 3%; 8% discontinued treatment because of adverse events. The rates were numerically lower than those reported for intravenous RYBREVANT plus LAZCLUZE in the Phase 3 MARIPOSA study. Separately, final overall-survival data from the Phase 3 PAPILLON study showed that intravenous RYBREVANT plus chemotherapy extended median overall survival to 34.3 months, versus 27.9 months with chemotherapy alone, in patients with advanced NSCLC carrying EGFR exon 20 insertion mutations. An analysis accounting for crossover by 76% of chemotherapy patients indicated a 43% reduction in the risk of death, while progression-free survival through a second disease progression reached 28.3 months versus 17.5 months. Johnson & Johnson shares were down 0.74% at $264.35 at the time of publication, according to Benzinga Pro data.
