Johnson & Johnson reported five-year follow-up data for CARVYKTI (ciltacabtagene autoleucel, or cilta-cel) in 20 patients with relapsed or refractory multiple myeloma who had received one to three prior lines of therapy. After a median follow-up of 60.7 months, 69.2% of patients were alive at five years, while 10 patients, or 50%, remained alive and progression-free after a single infusion without maintenance therapy. Median progression-free survival was 60.5 months, while median overall survival had not been reached in the previously reported analysis. All three patients assessed for minimal residual disease at five years were negative at the deepest tested level of 10^-6. The longer-term safety profile was consistent with known risks; one patient developed acute myeloid leukemia and two deaths occurred from progressive disease and new cancer. Separately, Tremfya met its primary and main secondary endpoints in the Phase 4 STAR trial among biologic-naive adults with active psoriatic arthritis and axial involvement. At Week 24, the IL-23 inhibitor improved composite disease activity scores, axial symptoms and sacroiliac joint inflammation versus placebo, with no new safety signals. Johnson & Johnson shares were down 0.20% at $270.24 at the time of publication, according to Benzinga Pro data.
